I have a hole in a stainless steel fermenter that is about 1 inch wide. What would be the best way to block/seal this off?
 
What you need is a "stainless steel weldless bulkhead". You'll have to search around to see if you can find one in the size you need.

If the one you find is tri-clamp on the outside then you'll also need a blanking plate, silicone gasket and clamp to complete the seal. If the one you find is threaded then you'll need a blanking plug and nylon washer. Or a ball valve tap, which you would turn off.
